Rational design, synthesis and study of new macrocyclic ligands for biomedical and radiopharmaceutical applications

The aim of the project is to understand the mechanisms that govern the thermodynamic and kinetic properties, both primordial for nuclear medicine applications, of the complexes formed with these novel hybrid ligands. Several hybrid ligands will be synthesized to study the thermodynamic, kinetic and structural properties of their complexes. Understanding the mechanism of complex formation allows the design of new molecules and improves their properties on a rational basis for a more efficient use in nuclear medicine. Combining this knowledge with previous data on rigid and preorganized macrocyclic ligands has the promise of faster complex formation with lanthanide (III) ions as compared to classical macrocyclic systems. By including such structural fragments in hybrid ligands, new ligand classes will be designed and synthesized, which may display fast formation kinetics.
